A Once-Daily Pill Shows Promise: Up to 12% Weight Loss in 36 Weeks
Imagine losing more than 12% of your body weight—without a single injection. That's the early promise of aleniglipron, a new GLP-1 pill that could shake up obesity treatment as we know it.
In a phase II clinical trial published in Nature Medicine, adults struggling with obesity or excess weight lost up to 12.1% of their body weight in just 36 weeks on this once-a-day pill. That’s not just a number—it’s the kind of result that could mean ditching the needle for a simple, daily tablet.
Aleniglipron works the same way as popular GLP-1 injectables like Wegovy and Ozempic, but here’s the twist: it’s a small molecule, not a peptide. In plain English, that means it’s a chemically made drug you swallow—no refrigeration, no needles, no complicated instructions. “Most meds you take—think aspirin or blood pressure pills—are small molecules,” explains Dr. Robert Kushner, one of the study’s authors and a longtime leader in obesity research. “You can take them with or without food, and even combine them with other drugs if needed.”
The study enrolled 230 adults, with an average age of 50, across 38 medical centers in the U.S. They were randomly assigned to one of three doses—45, 90, or 120 milligrams—or to a placebo. Over 36 weeks, those on the highest dose lost an average of 12.1% of their starting weight. Even the lowest dose produced a 9% drop. The placebo group, by comparison, saw just a 0.5% decrease.
As for side effects, most were mild to moderate stomach issues that became less common over time. About 1 in 10 participants stopped the treatment early, but no one developed liver problems linked to the drug.
Dr. Kushner says the results are a green light to move to larger phase III trials, with plans to slow dose increases to make the medication even easier to tolerate. “We didn’t spot any red flags or new safety concerns,” he says. “This could be a real step forward in how we treat obesity.”
If aleniglipron continues to shine, it could become a much more accessible option for millions of people—no prescription refrigeration, no injections, just a daily pill that fits right into your routine.
